刘德培课题组简介
本课题组从事基因表达调控与基因治疗、心血管疾病发病机制和重大疾病共性机制研究。承担国家自然科学基金重大、重点项目、科技部重大科技专项、中国医学科学院医学与健康科技创新工程等项目。课题组坚持以人为本,鼓励创新,充分发挥每个人的聪明才智,大家以诚相见,做到和而不同,为而不争,在这种宽松的学术环境中从事科学研究。
课题组负责人介绍
刘德培,男,研究员,博士生导师。1950年5月生,1986年获中国协和医科大学博士学位,1987-1990年美国加州大学旧金山分校博士后,1990年回协和基础所工作,1991年被授予“做出突出贡献的中国博士学位获得者”称号,1993年被评为“全国首届中青年医学科技之星”,1994年被评为国家级有突出贡献的中青年专家,入选原国家教委“跨世纪优秀人才培养计划”,获国家教委跨世纪优秀人才基金,获国家自然科学基金委优秀中青年人才基金,1995年获国家自然科学基金委杰出青年科学基金,被评为求是基金会杰出青年学者。1996年当选中国工程院院士,2008年当选为美国医学科学院(NAM)与第三世界科学院(TWAS)院士,2009年当选为欧洲科学院(EAS),2020年当选为中国中医科学院学部委员。现任重大疾病共性机制研究全国重点实验室主任,细胞生态海河实验室主任,中国医学科学院医学表观遗传学研究中心主任,中国医学科学院生命伦理学研究中心主任,国家人口健康科学数据中心主任。2016-2022年任国际医学科学院(IAPH)组织共同主席。国家自然科学基金委“优秀创新群体”学术带头人,心血管973项目首席科学家。获得卫生部科技进步奖、北京市科技进步奖、中华医学科技奖、国家自然科学奖。

陈厚早:男,研究员。北京协和医学院生化与分子生物学专业博士。2011年任中国医学科学院基础医学研究所研究员,2013年破格为博士生导师。获 “国家自然科学基金委优青” 、“中组部青年拔尖人才”,“科技创新领军人才”,“卫生部有突出贡献的中青年专家”,“国家自然科学基金委杰青”等荣誉或资助。现任中国生化与分子生物学脂质与脂蛋白青年委员会副主任委员,中国老年学和老年医学学会抗衰老委员会委员等。

课题组研究成果
在基因表达调控与基因治疗研究中,发现β珠蛋白基因簇红系增强子HS2及其关键位点,建立BAC介导的α/β珠蛋白基因簇转基因动物模型,发现α-基因簇活性染色质构象(ACH)中珠蛋白基因与看家基因共用转录工厂,发现β基因簇MAR间协同染色质高级构象,发现马利兰可显著增强贫血恒河猴与地贫患儿的胎儿型珠蛋白表达,用杂合寡核苷酸实现了β基因的定点修复并提出复制叉渗漏寡核苷酸掺入假说,发现Sirt1调节成纤维细胞向iPS细胞转化的效率,利用TALEN和CRISPR/Cas9技术结合同源重组修复病人来源的iPS细胞中的globin基因突变并进行两种方法的比较。
在心血管疾病发病机制研究中,建立ApoAI/CIII/AIV载脂蛋白基因簇转基因动物模型并发现ApoCIII增强子的调控机制;发现对氧磷酶(PON)基因簇能抑制动脉粥样硬化斑块形成并稳定斑块;发现内皮特异的III类去乙酰化酶Sirt1能降低ApoE敲除鼠的动脉粥样硬化,能量限制升高Sirt1的表达,抑制腹主动脉瘤的发生与严重程度,而衰老抑制Sirt1的表达,促进主动脉瘤的发生;发现Sirt2抑制衰老相关的心肌肥厚;发现Sirt4通过Sirt3促进心肌肥厚的发生;发现SIRT6 通过抑制炎症和衰老来抑制胸主动脉瘤;发现Echs1基因对于组蛋白巴豆酰化和心肌肥厚的影响;阐述氧化应激对于CLOCK修饰和生物节律的调控作用。发表综述阐述衰老和衰老相关疾病的四层作用机制。
以上工作在Circulation; European Heart Journal; Journal of Experimental Medicine; Circulation Research; PNAS; Nucleic Acids Research; Nature Cell Biology; STTT等杂志发表。共发表SCI论文200多篇,被引用超过10,000次。
